COMPARISON OF AEROSOL PERFORMANCE OUTPUT FROM ANTI-STATIC VALVE HOLDING CHAMBERS USING HFA METERED DOSE INHALER FORMULATIONS

Background: The valved holding chamber (VHC) has been developed to optimize aerosol delivery to patients. New VHCs are made of polymers having electrostatic dissipative (ESD) properties. A new ESD VHC has been developed and the in vitro aerosol characteristics have been compared to those of a currently available ESD VHC. Two HFA pressurized meter dose inhalers (pMDIs) - albuterol (90 μg/actuation, GlaxoSmithKline, Research Triangle Park, NC) and fluticasone (110 μg/actuation, GlaxoSmithKline, Research Triangle Park, NC) - were used in an in vitro test setup composed of a Next Generation Impactor (NGI, MSP Corp., St. Paul, MN) connected to a vacuum pump with flow rate set to 30 L/min.

Methods: The two VHC brands - a pre-production OptiChamber Advantage II (OCAII, Respironics, Inc., Respiratory Drug Delivery, Parsippany, NJ) and an AeroChamber Z-Stat (ACZ, Monaghan Medical Corp., Plattsburgh, NY) were pre-washed and air-dried prior to testing. The pMDIs were primed as per the instructions for use. The pMDI canisters were shaken and then fired 10 times into the NGI. Three units of each brand were tested, and each pMDI - VHC combination was tested in triplicate. The drug deposited in the NGI was analyzed by high performance liquid chromatography. The fine particle dose (FPD <4 �m) and the mass median aerodynamic diameter (MMAD) were calculated.

Results: There was no statistical significant difference in FPD or MMAD at p< 0.02. The FPD values (�g) for the VHCs with albuterol and fluticasone were respectively: OCAII=43.44, ACZ=42.87; OCAII=70.55, ACZ=66.42. The MMAD values (�m) for the VHCs with albuterol and fluticasone were respectively: OCAII=2.27, ACZ=2.29; OCAII=2.57, ACZ=2.56.

Conclusion: The aerosol characteristics are substantially equivalent between the OptiChamber Advantage II and the AeroChamber Z-Stat tested.
